Pulmonary vasodilation and hypotension in anaesthetised sheep following intratracheal administration of nitric oxide donors. The effect of intratracheal administration of nitric oxide (NO) donors on arterial blood pressure, pulmonary vascular resistance and pulmonary capillary wedge pressure (PCWP) was investigated in halothane anaesthetised ewes. The NO donors, S-nitroso-N-acetylpenicillamine (SNAP), S-nitroso-N-acetyl-D,L-penicillamine (SNAP-D,L-Pen) and 3-morpholinosydnonimine (SIN-1), were given at three different doses (i.e. 1, 5 or 10 mg/kg bodyweight) via the intratracheal route. While the dose of 1 mg/kg had no significant effect, the doses of 5 and 10 mg/kg produced a fall in mean arterial blood pressure which lasted 1-2 min. The maximum fall in mean arterial blood pressure after the administration of 10 mg/kg SNAP-D,L-Pen (46 +/- 8 mmHg) was less than after SNAP (73 +/- 7 mmHg) or SIN-1 (78 +/- 3 mmHg), however, the maximum fall in PCWP (24 +/- 2 mmHg) was similar in all three groups. The fall in mean arterial blood pressure correlated with the fall in PCWP in the two SNAP groups. The maximum fall in mean arterial blood pressure following SIN-1 (26 +/- 3 mmHg) was less than that following the two SNAPs. In anaesthetised sheep, intratracheal administration of a NO donor produced a fall in mean arterial blood pressure and in PCWP, the latter reflecting the fall in pulmonary vascular resistance. The response to SIN-1 appeared to be less than the other NO donors due to a smaller fall in PCWP.�
